Even exploring the environments on my third playthrough was enjoyable because of the many hidden nooks and paths to rediscover, and the multiple ways areas connect makes it easy to get lost again, but in a good way. As you explore, there are plenty of slimes to capture, scattered messages from the previous owner of the ranch to read, and a plethora of hidden chests to find. It’s a colorful world with diverse environments and cheery, whimsical music by Harry Mack that does a great job of adding magic and mysteriousness to some of the areas. But word was that Switch’s processor couldn’t handle the physics of the game, so I was worried that this version would have noticeable frame rate reduction, lower quality graphics, or even worse issues. But where Up sets its main character up for a journey to let go of his past and move on, Henry’s story is not so straightforward. The formation and maturation of VTCs requires the sequential action of two cytosolic coat protein complexes called COPII and COPI ( Aridor and Balch, 1995). In agreement with their function, VTCs localize both at the cell periphery ( Hauri et al., 2000 Klumperman et al., 1998) and in the juxtanuclear region at the cis-face of the Golgi complex. In mammalian cells, long-range transport of cargo proteins from peripheral ER exit sites (ERES) to the juxta-nuclear Golgi complex is mediated by pleiomorphic transport carriers called vesicular tubular clusters (VTCs) ( Bannykh and Balch, 1997 Lee et al., 2004 Tang et al., 2005). These observations strongly suggest that GBF1 regulates COPI membrane recruitment in the early secretory pathway. Strikingly, microinjection of anti-GBF1 antibodies specifically caused dissociation of COPI from membranes. Furthermore, recruitment of GBF1 to cargo-containing peripheral puncta coincided with recruitment of COPI, but not COPII. BrefeldinA-induced accumulation of GBF1 coincided with loss of COPI from peripheral puncta. Measurement of diffusion coefficients and subcellular fractionation confirmed this shift in GBF1 from cytosolic to membrane bound. Treatment with BrefeldinA dramatically altered this rapid exchange, causing accumulation of GBF1 on both Golgi and peripheral puncta before eventual redistribution to the ER in a microtubule-dependent manner. Live cell imaging revealed that GFP-GBF1 associates dynamically with both membranes through rapid exchange with a large cytosolic pool. Using new antisera and a GFP-tagged protein, we demonstrate that the exchange factor GBF1 localized to both Golgi membranes and peripheral puncta, near but separate from ER exit sites. Recruitment of COPI to nascent carriers requires activation of ADP-ribosylation factors by a BrefeldinA-sensitive guanine nucleotide exchange factor.
